Performance issue what to look for

Users are experiencing very slow response times when
-Posting invoices (SI's or PI's);
-Adding Lines to an SO;
-Opening Item/customer lists
I've tried to look for a pattern but it can happen at any time of the day.
Running table optimisation helped slightly
What can I do to pinpoint the cause of the issue?

Different things.
Might worth to restart the server.
Check sql server how much memory has been assigned to sql server
How many users are using , does server cope with all the users?
check hard disk IO on server is it fully busy? if yes then there might be activity running that you might not awrare of in the background.
