I just want to verify if modification on the header and line is still possible for closed/completed PO?

What do you mean by closed/completed PO?
Completely shipped and Invoiced?
if yes, it will be deleted from the database (in standard database)

As I said, it will be delted once you have shipped and invoiced completely.
You need to post Creditmemo and re-post PO with correct details..

If you invoice via the get receipts functionality then the order will not be automatically deleted in standard NAV. In this scenario yes you can reopen the purchase order and make amendments.
