the server service has to run under an account that has rights on the database.
If not then this error appears:
Either give system enough rights under the db or change the user of the service.

It's not the database that I'm using I change it in customsettings file and even restart all the services but it didn't work, still demanding database demo 6-0 or my DB name is Nav2009R2-DEMO.
I've also looked at the rights as you said and everything seems allright, do you have any idea what can be the cause?

make sure the database is sql and compatible with rtc (file -database - alter)
next make sure the correct customsettings.config is modified
check if the license is correct
check that you don't have multiple server services running.
