Is it possible to allow Item shipped first.
Subsequently follow up with Assigning Lot Number to the Shipments entry?

Physically is possible to ship the goods but NAV doesn't allow to ship the item without lot number and item tracking.

You can't do like that. NAV doesn't allow to do it.

Can i set Lot No not compulsory. Let user ship first.
Later revisit the Unposted Shipment to assign the Lot No.

By default, you can't achieve this. Little Workaround and customisation allows you to get your expected result.
