A Stock Keeping Unit works alongside items in the item table; it allows you to set different parameters for the same item at different locations - such as replenishment system, planning, cost, etc. For example, an item can have a replenishment system of 'purchase' at one location (head office) but other locations don't want to purchase the item, they want to transfer them from the head office when needed. So, a SKU card is set up for the other locations with a replenishment system of transfer, and the planning system takes this into account.
Responsibility centres don't specifically relate to stock. They allow you to set up an organisational unit (e.g. a sales office) and only allow people in that unit to see documents (sales / purchases) that relate to them - this might apply to geographical areas (e.g. only allow people in the north to see northern customers, and sales orders for those customers), or other organisational types - e.g. product range, etc.
