Windows Auth Fails

Windows Auth is failing with invalid username and password error on a 5.01 test database.
The same users can access a 4.03 db on the same server. I have checked SQL permissions and even the network admin gets this error.
When we synchronize permissions we get an hour glass and then it goes away.
What are the steps to troubleshoot this?

Does this file differ between 4.0 and 5.0?0

I couldn't tell you what the differences are, but at least comparing the file sizes between 4.0 SP1 and 5.0 SP1 the one for 5.0 is about 10% bigger. So some changes were probably made.0

Yes, and the newer version is backward compatible with older version.0
