Still I am getting the same error even after changing the firewall setting in Windows Firewall->Advanced Tab and changing Lan Area Connection--> Advanced setting and ticking Internet Mail server (SMTP).
First of all: can you telnet to the SMTP port on the mailserver (try telnet <servername> 25 from start->run). If you can, don't bother with firewalls, if you can't, there is still probably some other software blocking the port, like antivirus software (I know mcafee enterprise by defaults blocks port 25)
It's the default port for sending mail using SMTP. You won't find many servers which use another port to accept SMTP mail. Sending mail through SMTP is safe. It's blocked by some software because some malicious software also sends e-mail. Know all the spam you're receiving? lot's of these mails are send by spambots on computers which it's owner even doesn't know it does this.
If you're sure you don't have this kind of software on your computer (which should be detected by antivirus software and the likes) it's perfectly safe to open up the port - from the inside to the outside. You won't be receiving mail through SMTP, so the other way around is not necesarry
To be able for your server to receive e-mail through SMTP, port 25 needs to be opened on the server. It probably is, otherwise you would not receive any mail at all (assuming you don't use a POPreader). Check with your systemadministrator if port 25 is opened on the Exchange server. He can test it quite easily by telnetting on the server to localhost 25

Then how safe is mailing through SMTP protocol, if the port has been blocked by Antivirus.

Ours is a office setup which is using Microsoft Exchange Server in that case where the port no. 25 to be opened.
